OXIDATION OF OXYHEMOGLOBIN BY SODIUM NITRITE A kinetical study on the oxidation of hemoglobin to methemoglobin by nitrite

چکیده
The detailed mechanism of the reaction was further clarified by Martin et al4) in 1963 by investigating the relationship between the molecular structure of hemoglobin and the rate of the oxidation by nitrite. In these studies, however, the significance of the initial phase of the reaction, which will be clarified in the present paper, was insufficiently understood. As shown in Fig. 1, two phases, which were named lag and log phase in the paper, were observed in the reaction process. The properties of the lag and log phase was elucidated by carrying out the following experiments.

The kinetic differences between sodium nitrite, amyl nitrite and nitroglycerin oxidation of hemoglobin.
The effect of sodium nitrite, amyl nitrite and nitroglycerin (glyceryl trinitrate) on the hemoglobin of adult erythrocytes was examined in vitro. Both amyl nitrite and nitroglycerin reacted immediately with oxyhemoglobin to effect oxidation into methemoglobin while sodium nitrite required an inductionary period (lag phase) prior to the reaction. Kinetic studies of the biomolecular rate law for ...

Hemoglobin has functions besides carrying oxygen to the tissues, and regulates vascular tone and inflammation via a redox couple with methemoglobin. Hemoglobin has iron in the reduced valance Fe(II) and methemoglobin has iron in the oxidized valance Fe (III), with a free energy capable of producing water from oxygen. In generating methemoglobin the couple functions as a nitrite reductase. The d...
متن کاملKinetics and mechanism of the oxidation of human deoxyhemoglobin by nitrites.
The reactions of human hemoglobin with sodium nitrite and ethyl nitrite in deoxygenated media have been examined in kinetic detail. Nitrous acid has been identified from kinetic pH dependence studies as the principal oxidant of hemoglobin in reactions with sodium nitrite. Nitrosylhemoglobin is produced concurrently with methemoglobin as a result of reductive release of nitric oxide from nitrous...
متن کاملMechanism of interference by hemoglobin in the determination of total bilirubin. II. Method of Jendrassik-Grof.
Oxyhemoglobin in erythrocyte hemolysates interferes with the Jendrassik-Grof assay. Destruction of azobilirubin occurs when oxyhemoglobin is oxidized to methemoglobin during diazotization or to alkaline hematin with addition of alkaline tartrate. The most probable mechanism is by oxidation with an agent such as hydrogen peroxide or a related species resulting from hemoglobin oxidation. Methemog...
متن کاملNitrite uptake and metabolism and oxidant stress in human erythrocytes.
Nitric oxide, when released into the bloodstream, is quickly scavenged by Hb in erythrocytes or oxidized to nitrite. Nitrite can also enter erythrocytes and oxidize Hb. The goals of this work were to determine the mechanism of erythrocyte nitrite uptake and whether this uptake causes oxidant stress in these cells. Erythrocytes took up 0.8 mM nitrite with a half-time of 11 min. Nitrite uptake wa...
